摘要:
目的 探讨磷脂型 DHA( DHA-PC) 的抗肿瘤生长作用。
方法 :

应用超临界 CO2 萃取脱除胆固醇技术从鸢乌贼卵中提取 DHA-PC。采用 MTT 法、AO /EB 荧光染色法和流式细胞术研究 DHA-PC 对人肝癌 HepG-2 细胞增殖、凋亡的影响, 并进一步利用小鼠 S180 移植性肿瘤模型, 验证 DHAPC 的体内抑瘤效应。
结果 :

DHA-PC 能够抑制 HepG-2 细胞的增殖( P < 0. 05, P < 0. 01) , 诱导细胞凋亡, 干扰细胞周期, 使细胞停滞在 G0 /G1 期; 明显抑制荷瘤小鼠肿瘤的生长,其平均抑瘤率达 40. 94% 。结论 DHA-PC 在体内和体外都对肿瘤的生长具有抑制作用。
Methods:
DHA-PC separeted from cuttlefish spawn by supercritical carbon dioxide was used to prepare unilayer liposomes. The in vitro anti-cancer effect of DHA-PC was evaluated in human hepatocellular carcinoma HepG-2 cell lines by MTT method. Cell cycle and morphological features of HepG-2 cell were observed by flow cytometery and AO /EB staining, respectively. Moreover,The effects of DHA-PC on anti-tumor in vivo were investigated in transplanted tumor-bearing mice.

Results:
DHA-PC could effectively inhibit the proliferation of HepG-2 cell( P<0. 05,P<0. 01) ,induce apoptosis of tumor cells and block the HepG-2 cell at G0 /G1 phase; In vivo DHA-PC could inhibit the growth of tumor in mice, with an average inhibitory rate of
40. 94%.
Conclusion:
ZDHA-PC has strong anti -tumor activity both in vitro and in vivo and is a potential source of natural anti-tumor products.
